Human peripheral blood B cells originate from the bone marrow and are heterogeneous. The Epstein-Barr virus (EBV) can infect resting (naive) B cells in vitro, utilizing the gene products expressed during its latent phase to alter the B cell life cycle, thereby generating immortalized B lymphoblastoid cell lines (B-LCLs). This has become an effective and commonly used method for B cell immortalization in recent years. B-LCLs are often used as substitutes for peripheral blood B cells and as sources of DNA. Owing to their ability to secrete monoclonal antibodies, function in antigen presentation, and maintain a relatively stable genetic background, they are widely applied across various fields of life sciences.

Human B-lymphoblastoid cell lines (B-LCLs) are immortalized B lymphocyte cell lines cultured in vitro following EBV infection of B cells, capable of unlimited proliferation in culture.
